does the clock start ticking after you brew, the home brew, or is it already ticking in the box, I'm assuming the former? do glass jars have a better seal than plastic?

Home Brew will only need to be used when mixed. And then you have years of life in it. The glass jars are fractionally more airtight due to wadding.
